The effect of rotenone on the regulation of fatty acid synthesis in heart mitochondria.

چکیده
Acetate-lJ*C incorporation into long chain fatty acids by rabbit heart mitochondria was increased up to ZO-fold by citrate, isocitrate, malate, or succinate in the presence of rotenone and ATP. Rotenone blocked NADH oxidation and enabled the maintenance of a high NADH:NAD+ ratio. The rate of fatty acid synthesis was linearly related to the resulting endogenous mitochondrial NADH : NAD+ ratio. By inhibiting reversed electron flow, rotenone inhibited the early high rate of acetate incorporation observed with succinate oxidation. In this situation, succinate supplied reducing equivalents for fatty acid synthesis via malate dehydrogenase. Rotenone also blocked the ADPor dinitrophenol-induced cleavage of newly incorporated acetate-lJ*C from mitochondrial fatty acids by preventing a decrease in the NADH: NAD+ ratio. The NAD oxidation-reduction state is concluded to be the regulator of the rate and direction of the heart mitochondrial fatty acid synthetic-oxidation system.

متن کاملReduction pathway of cis-5 unsaturated fatty acids in intact rat-liver and rat-heart mitochondria: assessment with stable-isotype-labelled substrates.
Besides the conventional isomerase-mediated pathway, unsaturated fatty acids with old-numbered double bonds are also metabolized by reduction pathways with NADPH as cofactor. The relative contributions of these pathways were measured in intact rat-liver and rat-heart mitochondria with a novel stable isotope tracer technique. A mixture of equal amounts of unlabelled cis-5-enoyl-CoA and 13C4-labe...

Introduction & Objective: The link between dietary fat and coronary heart disease has attracted much attention since the effect of long?chain fatty acids (LCFA) on gene transcription has been established, which in part, these effects can be explained by the regulation of gene transcription. In this study, the P19CL6 cardiac cell?line was targeted for the investigation of (i) the effects of long...
متن کاملFusion of the Endoplasmic Reticulum and Mitochondrial Outer Membrane in Rats Brown Adipose Tissue: Activation of Thermogenesis by Ca2+
Brown adipose tissue (BAT) mitochondria thermogenesis is regulated by uncoupling protein 1 (UCP 1), GDP and fatty acids. In this report, we observed fusion of the endoplasmic reticulum (ER) membrane with the mitochondrial outer membrane of rats BAT. Ca(2+)-ATPase (SERCA 1) was identified by immunoelectron microscopy in both ER and mitochondria. This finding led us to test the Ca(2+) effect in B...
متن کاملProtective effect of metformin on toxicity of butyric acid and arsenic in isolated liver mitochondria and langerhans islets in male mice: an in vitro study
Objective(s): Arsenic, a toxic metal in drinking water and butyric acid (BA) is a free fatty acid found in many foods. These two can induce oxidative stress in some tissues. The present study investigated the protective effect of metformin against toxicity induced by Arsenic (As) and BA in isolated mice liver mitochondria and pancreatic islets. Materials and Methods: In this study, liver mitoch...
